The protein used in process of transformation was green fluorescent protein. Two tubes were labeled “+pGlo” and “-pGlo” and 250 μl of CaCl was added and tubes were placed on ice. I used a sterile loop and extracted 3 large circular colonies of E. coli then swirled it into the +pGlo tube and repeated this process for –pGlo tube. Under UV light both tubes displayed no color difference. The tubes were placed on ice for 10 minutes then heat shocked at 42 ˚C for exactly 50 seconds in order for transformation to occur with no damage to the cells. The tubes were immediately placed on ice for another 2 minutes before 250 μl of LB broth was added to each tube. An agar plate that contained Lb and amp a 100 μl of +pGlo suspension was added to the plate
