Mice will be exposed to Mn for several weeks through intratracheal administration. Animals will be subject to neurobehavioral tests for anxiety, motor and memory functions using plus maze apparatus, rotarod and Barnes maze tests respectively. Due to increased Mn uptake in Hfe-deficient mice, I expect to observe an impairment in neurobehavior after intratracheal exposure to Mn nanoparticles.
Specific aim 4: To investigate alterations in neurotransmitter proteins, neurotransmitter levels and oxidative stress markers due to the Hfe effect on Mn neurotoxicity.
Tissues from Specific Aim 3 will be used to determine the levels of isoprostane, an oxidative stress marker, using ELISA. Levels of neurotransmitter transporters and receptors will be measured
